In Mexico, flow cytometry has acquired great relevance in the agroindustrial, agricultural, forestry and livestock sectors, due to its capacity to analyze multiple characteristics of individual cells or heterogeneous populations.The objective of this work was to infer the ploidy level of 57 ecotypes of Bouteloua curtipendula (Michx.) Torr. from the absolute nuclear DNA content using leaves and a flow cytometry-based method. Leaves were processed with a simple mincing technique in a buffer solution to obtain cell nuclei in suspension, which were stained with propidium iodide and then analyzed with an Attune(R) acoustic focusing cytometer (Applied Biosystems) of the Flow Cytometry area of the National Laboratory for Agri-Food and Forestry Research and Service (LANISAF). For this purpose, leaf tissue nuclei from Zea mays L. were used as reference standard with DNA value 2C=5.67 pg.
