Design and Modeling of Peptide Competitive Inhibitors of HMG-Coenzyme A Reductase

Food-derived peptides represent a promising area of research due to their potential health benefits. In particular, peptides with hypercholesterolemic activity have attracted considerable attention owing to their ability to modulate elevated cholesterol levels, a major risk factor for cardiovascular diseases. Statins remain the most effective and widely used agents for the treatment of hypercholesterolemia. These compounds act as competitive inhibitors of 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR) and exhibit activity in the nanomolar range. This study reviews the methodologies employed to identify food-derived peptides acting as competitive HMGR inhibitors, as well as the design strategies used to enhance their bioactivity. To date, 36 such peptides have been reported. Comparative analysis of peptide design approaches revealed that strategies based on the relationship between conformational flexibility and bioactivity enable the rational design of novel peptides with nanomolar activity. Notably, the most active designed peptide demonstrated a 700-fold increase in activity compared to peptides naturally occurring in food. These findings highlight the potential of rational design approaches for the discovery of more potent peptides derived from natural proteins, with promising applications as nutraceuticals and therapeutic agents for the prevention of hypercholesterolemia.
